Currently there is no plugins mechanism for additional checks in amavisd-new, you will have to hack the code, probably somewhere in sub check_mail. I'm not sure a content filter is a proper place to implement disk quotas. These should be implemented by a local delivery agent, or perhaps by an external policy delegation filter in Postfix.
